Eloralintide

Eloralintide is an experimental, laboratory-made hormone analog developed by a pharmaceutical company as a candidate obesity treatment. It is designed to switch on one specific receptor for amylin, a hormone tied to feeling full after eating, while largely leaving related receptors alone. It remains in company-sponsored clinical trials and has not been approved anywhere; the trial data published so far describe body-weight changes in adults with obesity.

What it is

Eloralintide is a modified, once-weekly analog of human amylin built by a pharmaceutical company as a candidate treatment for obesity. Amylin is a natural hormone that signals fullness after a meal. Related experimental compounds activate more than one amylin-family receptor at once, but eloralintide is engineered to be far more selective for a single receptor type, called AMY1R, and researchers have proposed that this selectivity is behind its improved digestive tolerability compared with less selective amylin-receptor compounds.

The molecule is a modified 37-amino-acid chain with a fatty-acid side chain attached, a common engineering trick used to slow how quickly a peptide clears from the bloodstream so it can be given roughly once a week instead of daily. It remains an investigational compound: it has not been approved for any use in any country and is only available inside company-sponsored clinical trials.

How it is studied

Eloralintide has moved through the standard drug-development sequence: animal studies in obese rodents, a small early human safety study in healthy volunteers, and a larger, randomized, placebo-controlled trial in adults with obesity that ran for close to a year. The company that developed it has stated an intention to begin larger, later-stage human trials, including studies that combine it with another investigational weight-loss compound.

What studies report

  • In a multicenter, randomized, double-blind, placebo-controlled trial of 263 adults with obesity that ran 48 weeks, participants receiving eloralintide lost an average of 9.5 percent to 20.1 percent of their body weight depending on the amount given, compared with 0.4 percent in the placebo group, and the weight loss had not leveled off by the end of the trial [1][2]. What is not known

The 48-week trial is the only completed randomized human study reported for eloralintide so far, and it does not show what happens beyond that point: whether weight loss continues, plateaus, or reverses with longer treatment or after stopping is not yet known. The trial was conducted in adults with obesity; how the compound behaves in other groups, including people with other metabolic conditions, has not been reported. As an investigational compound, eloralintide is not available outside sponsored trials, so no independent, non-company-funded studies exist yet.

Eloralintide is an investigational drug candidate. It has not been approved by the United States Food and Drug Administration or by any other national medicines regulator, and it is not legally available outside company-sponsored clinical trials.
