Ovagen is a lab-made chain of three amino acids, sometimes called EDL after the initials of its building blocks. Russian researchers built it as part of a family of short peptides meant to nudge specific organs, and it is marketed in Russia as a food supplement for liver and digestive support. Yet nearly every indexed research paper on it studies kidney tissue in rats, not the liver, which makes it a useful example of how marketing language and published research can point in different directions.
What it is
Ovagen is built from just three amino acids in the sequence glutamic acid, aspartic acid, and leucine. It belongs to a class of short peptides researchers call bioregulators, developed at a St. Petersburg research institute and studied for effects that appear tied to specific organs rather than the whole body.
Laboratory work on Ovagen has focused on kidney cells and kidney injury in rats. Researchers propose that the peptide can enter a cell's nucleus and bind to DNA at a specific stretch of genetic code, which they think changes which genes are switched on. The authors of these studies describe the exact way it crosses the cell membrane to reach the nucleus as still unclear.
How it is studied
All of the primary evidence on Ovagen comes from rat studies: kidney cells grown in a dish (in vitro) and whole rats given the peptide before or after kidney injury caused by drugs or reduced blood flow. No human trials of Ovagen have been published in the sources reviewed here.
What studies report
- In rats given the chemotherapy drug cisplatin, which damages kidneys, Ovagen treatment brought urine output, a blood marker called creatinine, and kidney filtering ability closer to normal compared with untreated rats. [1]
- In rats whose kidneys were injured by the antibiotic gentamicin or by cutting off part of their blood supply, Ovagen treatment was linked to less protein in the urine, lower salt loss, and better antioxidant enzyme activity. [2]
- In kidney cells taken from rats and grown in culture, adding Ovagen increased cell growth and lowered several markers linked to cellular aging (p16, p21, p53), while a protein tied to cell repair, called SIRT-6, increased. [3]
- In old rats, Ovagen increased urine output and sodium excretion and raised antioxidant enzyme activity in kidney tissue, with no sign of kidney-damaging effects on tissue examination. [4]
- The researchers who found the DNA-binding effect propose that Ovagen and a related peptide bind a specific DNA sequence in a groove of the double helix, which they think is how the peptide changes which aging-related genes are active; this remains a proposed mechanism, not a directly observed one. What is not known
No published study has tested Ovagen's safety or effects in people, and nearly all of its primary research traces back to one laboratory, St. Petersburg Institute of Bioregulation and Gerontology, so there has been little independent replication. Its proposed DNA-binding mechanism rests on computer modeling of how the peptide might fit against a DNA sequence, not on a directly observed binding event inside a cell. The liver and digestive claims found in some marketing are not supported by any primary research paper identified for this entry, which focuses only on kidney research; readers should treat that gap as a reason for caution, not confirmation either way about liver effects.
Regulatory status
Ovagen is not approved as a drug by the FDA, the European Medicines Agency, or, based on the sources reviewed, any verified Russian pharmaceutical registration. In Russia it is marketed as a food supplement, a different regulatory category than a medicine, meaning it has not gone through the same clinical testing required for drug approval anywhere.
